Purpose:

To determine the maximum safe dose of intravitreal clonidine (IVC) in rabbits´ eye

Methods:

20 rabbits were divided into 3 groups (A to C). 5, 7 and 8 rabbits in groups A to C received 15, 25 and 50 micrograms (µg) of IVC in their right eyes, respectively. The left eye of the rabbits was considered as control and received 0.1ml of balanced salt solution (BSS). Electroretinogram (ERG), was obtained at baseline, also 1, 4 and 8 weeks after IVC injection in all groups. Afterwards the animals were euthanized and their eyes enucleated for routine histopathologic analysis.

Results:

ERG and histopathologic studies demonstrated no signs of retinal toxicity after IVC injection, except for significant reduction of scotopic mean b-wave amplitude which was detected in group C, 8 weeks after IVC injection compared to the baseline (P= 0.018).

Conclusion:

IVC with a dose of 25µg/0.1ml was found to be safe in rabbits´ eye. Further studies are needed to confirm these preliminary results and investigate the possible neuroprotective effects of clonidine.
